DNA Sequencing

The AGTC DNA Sequencing Lab provides fast, reliable and accurate DNA sequencing for a variety of sample types including plasmid DNA as well as PCR product. The Sequencing Lab is equipped with an Applied Biosystems ABI Prism 3730 DNA Analyzer, which is able to sequence as many as 96 samples per sequencing plate and over 570 samples per day. The DNA sequencing is accomplished using the ABI BigDye® Terminator v3.1 Cycle Sequencing Kit.  With the BigDye® Terminator v3.1 chemistry, you get robust, highly flexible chemistries for a wide range of applications, including de novo sequencing and resequencing.

We offer the investigator two options for sequencing: you can perform your own sequencing reactions with the ABI BigDye® Terminator v3.1 Cycle Sequencing Kit (will need to be purchased by researcher) or the reactions will be done by our staff using our ABI BigDye® Terminator v3.1 Cycle Sequencing Kit. The researcher can submit his/her own custom-designed primers (see primer concentrations for details) or use our common primers ((T7, T3, SP6, M13 Forward & M13 Reverse) provided by the Sequencing Lab at no additional charge. For required primer concentrations and quantities, please click here.

Submitting DNA samples for Sanger Sequencing:

Wayne State University now requires all DNA sequencing request to our facility to be submitted through the iLab Solutions website.  Please note, this is a University policy and we will not be able to process any request not submitted through this website.  The iLab Solutions website is located at: https://waynestate.corefacilities.org/

For Wayne State researcher, you will login using your Wayne Access ID and password - these are the same ones you use to access the WSU Academica.  If you work outside of Wayne State University, you will have to request an account from iLab solutions - this option is listed on the website.

After logging in, the AGTC home page should open, if not, you can choose it from the Core Facilities tab on the left side of the page.  Instruction manuals on how to use the iLab site are located in the Getting Started in iLab section.  You can download the sequencing request forms to be upload to iLabs here:  AGTC Sequencing Request or AGTC 96well Sequencing Request.

If you are requesting more than 33 sequences, it is less expensive to submit them in a 96-well plate format.  You can use either a 96-well PCR plate (do not use flat bottom culture plates) or you can use PCR strip tubes (you will need to clearly label these tubes).  You do not need to submit a full plate of 96 samples to use the AGTC 96well Sequencing Request form.

Sequencing results will be sent electronically to your email account. If requested, a paper hard-copy of the chromatogram can be sent for a nominal charge.
